4. Pharmacokinetics -> Well absorbed and distributed ! ->Accumulates in any fluid collection (ascites, pleural, pericardial effusion) ! ->Higher than expected toxicity due to prolonged elimination !
5. Pharmacodynamics -> Inhibits the action of the enzyme dihydrofolate reductase ! -> Results in cell death ! -> Folinic acid used in high-dose methotrexate therapy to prevent cell death
6. Pharmacotherapeutics -> Acute lymphoblastic leukemia ->Choriocarcinoma ->Osteogenic sarcoma ->Malignant lymphomas ->Cancers of the head, neck, bladder, testis, and breast
7. Unconventional Treatment -> Severe psoriasis -> Graft versus host disease -> Rheumatoid arthritis
8. Disease-Modifying Anti- Rheumatic Drug (DMARD) ->Decreases pain and swelling of arthritis -> Requires careful monitoring to reduce risk of liver injury and infections. ->Can cause serious birth defects
10. Side Effects of Methotrexate ->Dyspnea ->Headaches ->Liver cirrhosis ->Confusion and irritability -Bruising ->Skin sensitivity to the sun
