アンモニウム試験法は、医薬品に混在するアンモニウムイオンを測定する方法です。アンモニアを蒸留で捕え、インドフェノールの呈色を比較液と比べることで調べます。

  1. 日本薬局方－一般試験法 1.02 アンモニウム試験法 2020/6/1 Ver. 1.0

  2. アンモニウム試験法とは？ 医薬品中のアンモニウムイオン（NH 4 +）を測定する方法 • 蒸留によりアンモニウムを分離・回収する • 蒸留には大気圧、減圧の2つがある • 試薬の呈色を標準液と比較して調べる

  3. 蒸留装置 左が大気圧、右が減圧し蒸留するもの • 左の丸底フラスコに試料を入れる • 突沸防止用のガラス管を試料に漬け る • 減圧はアスピレーターで行う •

    減圧は60ºCで水浴、大気圧は5-7 mL/分となるよう温める • 採取側にはホウ酸水溶液を入れる

  4. 検体の試験法 フェノール・ペンタシアノニトロシル鉄(III)酸ナトリウム＊の 呈色で調べる • 30 mLをネスラー管に入れ、＊試液6 mLを加える • 次亜塩素酸ナトリウム・水酸化ナトリウム4 mL、水50mLを加える

    • 60分放置する • インドフェノールの青色呈色を比較液と比べる
